چکیده
background: considering that cannabinoids protect neurons against neurodegeneration, in this study, the neuroprotective effect of win55,212-2 in paraoxon induced neurotoxicity in pc12 cells and the role of the n-methyl-d-aspartate (nmda) receptor were evaluated. materials and methods: in this study pc12 cells were maintained in dulbecco's modified eagle’s medium (dmem+f12) culture medium supplemented with 10% fetal bovine serum. the cells were treated with paraoxon (200 μm) in the presence or absence of win55,212-2 (0.1 μm), nmda receptor agonist nmda (100 μm), cannabinoid receptor antagonist am251 and nmda receptor antagonist mk801 (1 μm) at 15 minutes intervals. after 48 hours of exposure, cellular viability and protein expression of the cb1 receptor were evaluated in pc12 cells. results: following the exposure of pc12 cells to paraoxon (200 μm), a reduction in cell survival and protein level of the cb1 receptor was observed (p<0.01). treatment of the cells with win55,212-2 (0.1 μm) and nmda (100 μm) prior to paraoxon exposure significantly elevated cell survival and protein level of the cb1 receptor (p<0.01). also, am251 (1μm) did not inhibit the cell survival and protein level of the cb1 receptor increase induced by win55,212-2 (p<0.001). however, mk801 (1 μm) did inhibit cell survival and protein expression of the cb1 receptor increase induced by nmda (p<0.001). conclusion: the results indicate that win55,212-2 and nmda protect pc12 cells against paraoxon induced toxicity. in addition, the neuroprotective effect of win55,212-2 and nmda was cannabinoid receptor-independent and nmda receptor dependent, respectively.

Background Anti-N-methyl- d-aspartate receptor (anti-NMDAR) encephalitis is an autoimmune neurological disorder that usually occurs as a paraneoplastic syndrome and is particularly associated with ovarian teratoma. Standard therapy for severe cases is not established and the prognosis in patients who do not respond to first-line treatment is poor. Case Report An 11-year-old boy complained ps...

Objective(s): -methyl-D-aspartate NMDA receptor (NMDAR) and aquaporin 4 (AQP4) are involved in the molecular cascade of edema after traumatic brain injury (TBI) and are potential targets of studies in pharmacology and medicine. However, their association and interactions are still unknown.Materials and Methods: We established a rat TBI model in this study. The cellular distribution patterns of ...
متن کاملThe effect of morphine dependence on expression of hippocampal N-methyl-D-aspartate receptor subunits in male rats
Introduction: N-methyl-D-aspartate (NMDA) receptors play a pivotal role in the development of tolerance and physical dependence to opiates. Activation of NMDA receptors involves the induction of long term potentiation (LTP) in hippocampus. Our previous study suggested that chronic oral administration of morphine enhanced NMDA dependent LTP in the CA1 area of hippocampal slices of rats. The p...
متن کاملBryonolic Acid, a Triterpenoid, Protect Against N-methyl-d-Aspartate-Induced Neurotoxicity in PC12 Cells.
Calcium overload is considered to be one of the mechanisms of cerebral ischemia. Ca(2+) influx and Ca(2+)/calmodulin-dependent protein kinase II (CaMKII) and cAMP response element-binding protein (CREB) phosphorylation are considered to be involved in N-Methyl-d-aspartate (NMDA)-induced apoptosis process. This study investigated the neuroprotective effects of bryonolic acid (BA) in an NMDA-indu...
متن کاملEffect of Cannabinoid Receptor Agonist WIN55, 212-2 on the Anxiety Induced by PTSD in Male Rats
Introduction: Posttraumatic stress disorder is a severe anxiety disorder caused by exposure to traumatic events. The aim of this study was to induce PTSD in rats and examine the effect of WIN55-212-2, a cannabinoid receptor agonist, on anxiety. Methods: SPS&S model was used to induce PTSD in 56 male Wistar rats. Rats were restrained for 2 h, immediately followed by forced swimming for 20 mi...
